针刺“四关”穴对阿尔茨海默病大鼠学习记忆及海马区β淀粉样蛋白42、白介素-1β和白介素-2的影响

摘要(Abstract):

目的:观察针刺"四关"穴对阿尔茨海默病(AD)大鼠海马区白介素-1β(IL-1β)、白介素-2(IL-2)及β淀粉样蛋白(Aβ)42表达的影响,探讨针刺"四关"穴对AD的作用机制。方法:将SD大鼠随机分为假手术组、模型组、西药组、针刺组,每组12只。采用侧脑室注射链脲佐菌素制备AD模型。针刺组取"四关"穴(双"合谷""太冲"),留针15min,间隔5min行针1次,平补平泻,频率为120~150次/min,捻转角度为90°~180°,治疗6d休息1d为一疗程,共4个疗程;西药组进行盐酸多奈哌齐混悬液(0.45mg/kg)灌胃治疗。应用Morris水迷宫测试各组大鼠学习记忆能力的变化,采用免疫组织化学方法检测大鼠海马Aβ42的表达情况,采用双抗体夹心法检测IL-1β以及IL-2在大鼠海马中的含量。结果:与假手术组比较,模型组大鼠平均逃避潜伏期延长(P<0.05),平均穿越平台次数减少(P<0.05),海马区Aβ42的阳性细胞表达和IL-1β的含量增多(P<0.05),IL-2含量下降(P<0.05);与模型组比较,西药组、针刺组大鼠平均逃避潜伏期明显缩短(P<0.05),平均穿越平台次数明显增加(P<0.05),海马区Aβ42阳性表达和IL-1β含量降低(P<0.05),IL-2含量升高;与西药组比较,针刺组大鼠平均逃避潜伏期和穿越平台的次数差异无统计学意义(P>0.05),而海马区Aβ42的阳性细胞表达和IL-1β含量降低(P<0.05),IL-2含量升高(P<0.05)。结论:针刺"四关"穴能一定程度上改善AD大鼠学习记忆能力,其机制可能与降低海马中IL-1β含量、升高IL-2含量,以及改善Aβ沉积有关,且作用效果与西药盐酸多奈哌齐相当。
Objective To observe the effect of acupuncture stimulation of bilateral"Hegu"(LI 4)and "Taichong"(LR 3,the so-called"Four Gate Points")on learning-memory ability,hippocampal interleukin-1(IL-1)βand IL-2and amyloidβ(Aβ)42levels in Alzheimer's disease(AD)rats,so as to reveal its underlying mechanism in improving AD.Methods Male SD rats were randomly divided into sham operation,model,medication and acupuncture groups(n=12rats in each group).The AD model was created by microinjection of streptozotocin(10μL,3mg/kg)into the lateral ventricle(repeated the microinjection once two days later).Bilateral LR 3and LI 4were punctured with filiform needles and stimulated manually,once a day,6days a week for 4weeks.The rats of the medication group were intragastric perfusion of Donepezil HCl(0.45mg/kg),once a day for 4weeks.The learning-memory ability was detected by Morris water maze swimming tests.The immunoactivity of hippocampal Aβ42was detected by immunohistochemistry,and the contents of IL-1βand IL-2in the hippocampus tissue were determined by ELISA.Results After modeling,the average escape latency of Morris water navigation task was significantly increased,and the target-platform crossing times of space probe trials were significantly reduced in the model group(P<0.05),suggesting a lowering of learning-memory ability.After acupuncture intervention,the increased escape latency and the decreased target-platform crossing times were reversed,suggesting an improvement of the learning-memory.The hippocampal Aβ42immunoactivity and IL-1βcontent were significantly higher in the model group than in the sham operation group(P<0.05),but the hippocampal IL-2content was markedly decreased in the model group(P<0.05).Following the interventions,the increased Aβ42expression and IL-1βcontents,and the decreased IL-2contents in the hippocampus were also reversed in both the acupuncture and medication groups(P<0.05).Conclusion Acupuncture may improve learning-memory ability in AD rats,which may be associated with its effects in reducing hippocampal Aβ42expression and IL-1βcontent and in up-regulating IL-2level.
