基于“肾脑相济”理论的电针疗法对痴呆小鼠脑内β-淀粉样蛋白及中性内肽酶表达的影响

摘要(Abstract):

目的:探讨基于"肾脑相济"理论的电针疗法对快速老化SAMP 8小鼠大脑颞叶皮层β-淀粉样蛋白(Aβ)含量及中性内肽酶(NEP)表达的影响,探讨电针治疗阿尔茨海默病(AD)的可能机制。方法:将8月龄雄性快速老化SAMP 8小鼠随机分为模型组、电针组、美金刚组,每组6只。使用同月龄正常老化SAMR 1小鼠6只作为对照组。电针组电针小鼠"百会""肾俞""太溪"穴,每次15min,每日1次,治疗8周,美金刚组予美金刚灌胃(20mg·kg-1·d-1),连续8周。采用ELISA法、免疫组织化学法、Western blot法观察小鼠大脑颞叶皮层内Aβ含量和NEP的表达水平。结果:与对照组比较,模型组小鼠大脑颞叶皮层内Aβ1-40和Aβ1-42含量明显增高,NEP表达水平明显下降(P<0.05);与模型组比较,电针组和美金刚组小鼠大脑颞叶皮层内Aβ1-40和Aβ1-42含量明显下降,NEP表达水平明显上升(P<0.05);电针组与美金刚组各指标比较差异无统计学意义(P>0.05)。结论:"肾脑相济"电针疗法对SAMP 8小鼠大脑颞叶皮层中NEP表达有较好的调控作用,能抑制脑内Aβ的生成,从而防治AD。
Objective To observe the effect of electroacupuncture(EA)on the levels of Aβ1-40 and Aβ1-42 and neprilysin(NEP)protein in the temporal cortex of SAMP 8 mice with Alzheimer's disease(AD),so as to explore its mechanism underlying improvement of AD based on the theory of"mutual assistance of kidney and brain"in traditional Chinese medicine.Methods Eighteen male SAMP 8 mice were randomly assigned to 3 groups:model,EA and medication(memantine,n=6/group),and 6 male homologous SAMR 1 mice were used as the normal control group.According to the"mutual assistance of kidney and brain"theory,EA(2 Hz,0.6 mA)was applied at"Baihui"(GV 20),bilateral"Shenshu"(BL 23)and"Taixi"(KI 3)for 15 min,once daily for8 consecutive weeks.Mice of the medication group were treated with intragastric administration of memantine(20 mg·kg-1·d-1)for 8 weeks.The contents of Aβ1-40 and Aβ1-42 in the temporal cortex were assayed using ELISA,and the expression of NEP protein in the temporal cortex was detected using immunohistochemistry and Western blot(WB),separately.Results The contents of Aβ1-40 and Aβ1-42 were significantly higher in the model group than in the control group(P<0.05),and notably lower in both EA and medication groups than in the model group(P<0.05).WB assay showed that the expression level of NEP was significantly down-regulated in the model group relevant to the control group(P<0.05),and obviously up-regulated in both EA and medication groups relevant to the model group(P<0.05).No significant differences were found between the EA and medication groups in the contents of Aβ1-40 and Aβ1-42 and in the up-regulated expression levels of NEP protein(P>0.05).Immunohistochemistry stain showed that NEP-positive products mainly expressed in the perikaryon of neurons of the temporal cortex.Conclusion EA intervention based on"mutual assistance of kidney and brain"theory has a good efficacy in up-regulating NEP expression and down-regulating Aβ1-40 and Aβ1-42 levels in the temporal cortex of AD mice,suggesting apotential effectiveness in the prevention and treatment of AD.
