针灸上调血清Aβ内化酶对阿尔茨海默病大鼠学习记忆能力和β淀粉样蛋白沉积的影响

摘要(Abstract):

目的:探讨针灸通过降解β淀粉样蛋白(Aβ)改善阿尔茨海默病(AD)大鼠学习记忆能力的作用机制。方法:Wistar大鼠随机分为正常组、假手术组、模型组和针灸组,每组10只。采用双侧海马注射5μL Aβ_(1-42)复制AD模型。针灸组"百会"及"肾俞"穴针刺加艾灸治疗,15min/次,1次/d,6d为1个疗程,治疗2个疗程。采用Morris水迷宫检测各组大鼠行为学,ELISA法检测血清Aβ_(1-42)及Aβ内化酶的含量,免疫组化法检测海马齿状回内Aβ_(1-42)表达变化。结果:与假手术组比较,模型组前5d及后3d平均逃避潜伏期明显延长,有效区停留时间明显缩短,跨越原平台位置的次数明显减少(P<0.01),血清中转甲状腺素蛋白(TTR)、脂蛋白酯酶(LPL)、α_2巨球蛋白(α_2M)及载脂蛋白E(ApoE)含量明显下降(P<0.01,P<0.05),Aβ_(1-42)的含量明显增多(P<0.01),齿状回内Aβ_(1-42)阳性表达升高(P<0.01)。与模型组比较,针灸组前5d及后3d平均逃避潜伏期明显缩短,有效区停留时间明显延长,跨越原平台位置的次数明显增多(P<0.01,P<0.05),血清中TTR、LPL、α_2M及ApoE含量明显上升(P<0.01,P<0.05),Aβ_(1-42)含量明显减少(P<0.05),齿状回内Aβ_(1-42)阳性表达降低(P<0.05)。结论:针灸"百会""肾俞"能明显改善AD大鼠学习记忆能力,其机制可能与上调血清中Aβ内化酶的含量,促进Aβ的清除以保护神经细胞有关。
Objective To observe the effect of acupuncture and moxibustion(AM)on learning-memory ability and expression of amyloid beta(Aβ)in the hippocampal dentate gyrus(DG)of Alzheimers disease(AD)rats,so as to explore its mechanism underlying improvement of AD. Method Forty male Wistar rats were randomly divided into normal,sham operation,model and AM groups(n=10 in each).The AD model was established by bilateral hippocampal injection of Aβ_(1-42)(5 μL).The AM was applied at"Baihui"(GV 20)and"Shenshu"(BL 23)for 15 min,once daily for 12 times.Morris water maze tests were used to assess the rats' learning-memory ability.The levels of serum Aβ_(1-42) and Aβinternalizing enzymes including transthyretin(TTR),lipoprotein lipase(LPL),alpha 2 macroglobulin(α_2 M)and apolipoprotein E(ApoE)were detected by ELISA.The expression of Aβ_(1-42) in the hippocampal DG was detected by immunohistochemistry.Results Compared with the sham operation group,the average escape latency of location navigation test was significantly prolonged in the first 5 days and the last 3 days(P<0.01),and the residence time of the target-quadrant and the platform crossing times of special probing task were significantly reduced in the model group(P<0.01).After the intervention,the average escape latency was obviously shortened in the first 5 days and the last 3 days(P<0.01),and both the residence time and the platform crossing times were significantly increased in the AM group relevant to the model group(P<0.01,P<0.05).After modeling,the contents of TTR,LPL,α_2 M and ApoE in the serum were significantly lower in the model group than in the sham operation group(P<0.01,P<0.05),while the levels of serum Aβ_(1-42) and hippocampal Aβ_(1-42) expression were considerably increased relevant to the sham operation group(P<0.01).Following the intervention,the TTR,LPL,α_2 M and ApoE contents in the serum were significantly increased(P<0.01,P<0.05),and both serum Aβ_(1-42) and hippocampal Aβ_(1-42) expression were notably down-regulated in comparison with the model group(P<0.05).No significant differences were found between the normal and sham operation groups in the levels of all the abovementioned indexes(P>0.05).Conclusion AM can improve the learning-memory ability of AD rats,which may be related to its effects in up-regulating the contents of serum Aβinternalizing enzymes and promoting the clearance of hippocampal Aβ.It suggests a protective role of AM on hippocampal neurons.
