累加电针对脑缺血大鼠皮层脑源性神经营养因子表达及脑梗塞体积的影响
Effect of Electroacupuncture on BDNF Expression at Ischemia Cortex and Infarct Volume after Middle Cerebral Artery Occlusion in Rat

摘要(Abstract):

在大脑中动脉阻塞 (MCAo)及再灌注模型上 ,观察缺血后一次电针或累加电针对大鼠皮层脑源性神经营养因子 (BDNF)表达和脑梗塞体积的影响。MCAo致缺血 90min ,一次电针在缺血后立即给予 ,累加电针组则每天给予电针 1次 ,电针取“水沟”和“百会”穴 ,时间为 1h。各组均在缺血再灌 7天后进行行为评分 ,取脑进行免疫组织化学染色和 2 ,3,5 三苯基氯化四氮唑 (TTC)染色。结果表明 ,累加电针组在缺血灶周围皮层表达的BDNF免疫阳性细胞数量多于一次电针和缺血组 (P <0 .0 5 ) ,同时功能恢复也优于一次电针。
The model of reversible middle cerebral artery occlusion (MCAo) was used to study effect of electroacupuncture (EA) on brain derived neurotrophic factor (BDNF) expression at ischemia cortex and infarct volume after MCAo in rats. MCA was occluded for 90 min and one hour EA was applied immediately at points of “Shuigou” and “Baihui” after cerebral ischemia. In accumulated EA group one hour EA was applied everyday till the rats were decapitated. After reperfusion 7 days, behaviour assessment ,immuohistochemistry and TTC staining were applied. Results suggested that BDNF immunoreactive cells expressed mainly in the peripheral cortex of the infarction. In accumulated EA group the expression of BDNF was higher than that in ischemia and EA group (P<0.05),and its behaviour assessment was the best.
